Calcium/calmodulin dependent protein kinase II gamma

Family: CAMK (Calcium/Calmodulin dependent kinase)

CaMK2G is the gamma chain of CaMK2 protein. CaM kinases are Ca2+/calmodulin dependent and play important part in the Ca2+ signal transduction pathway. Its α and β present at very high levels in brain and γ and δ isoforms present in heart, brain, and other tissues. CaMK2 is a key target in the cardiac field, it is a promising target in heart failure, arrhythmia, and other forms of heart disease 1. Few CaMK2 inhibitors were identified including KN-93, KN-62, which were shown to bind CaMKI, CaMKIV, ion channels as well. These inhibitors bind allosterically and do not inhibit autonomous kinase activity. Few peptides were shown to bind and inhibit CaMK2, providing higher selectivity. ATP-compeptive small molecule therapeutics were also developed including Scios 15b, SMP-114 and Bosutinib.
